Sildenafil Citrate
Indications
Sildenafil is suitable for the treatment of erectile dysfunction.
Pharmacology
Sildenafil is a selective inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5) and is used to treat erectile dysfunction. Danafil (sildenafil) enhances the effect of nitric oxide (NO) by inhibiting phosphodiesterase type 5 (PDE5), which is responsible for the degradation of cGMP in the cavernous body, leading to smooth muscle relaxation and blood flow to the sponge Body.
Dosage & Administration
The recommended dose of Sildenafil is 50 mg collected approximately one hour before sexual activity. However, Sildenafil can be taken from 30 hours to 4 hours of sexual activity. On the basis of effectiveness and tolerance, the dose can increase up to 100 mg or decrease at 25 mg. The frequency of maximum recommended administration is once a day.
Interaction
Simultaneous use of sildenafil and organic nitrates for angina pectoris can cause hypotension. Cimetidine, a drug used to treat gastric ulcers, some antibiotics such as erythromycin and rifampicin, and some protease inhibitors such as ritonavir and saquinavir, are used to treat HIV infection. Denafil increases plasma concentration. Some drugs used to treat fungal infections, such as ketoconazole and itraconazole, can reduce the clearance of sildenafil.
Contraindications
Sildenafil is contraindicated in patients who are allergic to any component of sildenafil. Sildenafil can enhance the antihypertensive effect of nitrate drugs, so patients who take organic nitrate drugs regularly or intermittently are contraindicated.
Side Effects
Side effects of sildenafil treatment include headache, flushing, indigestion, nasal congestion, urinary tract infection, abnormal vision, diarrhea, dizziness, and rash.
